HEPES is a commonly used zwitterionic buffer that can maintain a constant pH range for a long time. The effective buffer range is pH 6.8-8.2, and pKa (25°C) = 7.5. HEPES solution (1M, pH7.3) is commonly used in cell culture, and the general working concentration is 10-25mM. Compared with the low pKa bicarbonate buffer, HEPES has a pKa of 7.5 and has good buffering capacity in the pH range of 7.2-7.6, which is suitable for cell culture that requires precise pH conditions. Since the buffering capacity of HEPES is independent of CO2 concentration, HEPES can also provide good buffering capacity in open culture or when cells are out of the CO2 incubator for a long time (such as cell observation). HEPES is the preferred buffer for many cell culture systems because HEPES has no permeability to cell membranes, has little effect on various biochemical reactions, is chemically and enzymatically stable, and has very low visible and UV light absorption. HEPES is non-toxic to cells at a concentration of 10-25mM, but may be toxic to some cells and affect cell growth when the concentration exceeds 40mM. The HEPES solution (1M, pH7.3, for cell culture) produced by Biologix can be directly used for cell culture and other purposes after filtration and sterilization, and no filtration or other sterilization treatment is required before use.
